Buona conoscenza della chimica organica di base, stereochimica e reattività dei gruppi funzionali. Tecniche di sintesi, isolamento e purificazione, nozioni di base di Chimica Generale,Chimica Organica e Chimica Fisica.
Per la parte "Meccanismi" (G. Tonachini): Nozioni di base di chimica organica e chimica fisica, così come fornite dagli insegnamenti di chimica organica e chimica fisica del triennio.
Fundamentals of organic chemistry, stereochemistry and reactivity of functional groups. Simplemsynthetic sequences, purification and identification of simple compounds. Physical chemistry, such as thay are provided by the organic and physical chemistry courses of the first three years.

Obiettivi formativi

L'insegnamento si propone obiettivi distinti per la parte di sintesi e per la parte di studio dei meccanismi.

Per la parte di Sintesi (Docente Prandi):

Al termine del corso, lo studente è in grado di scegliere la metodologia più adatta per la formazione di legami carbonio-carbonio utilizzando composti organometallici (preparati preventivamente o generati in situ), complessi p di metalli di transizione,  con il massimo controllo di regioselettività, chemoselettività e stereoselettività. 

Parte "Meccanismi" (G. Tonachini):

L'obiettivo è di fornire semplici strumenti teorici atti a razionalizzare o a predire in termini qualitativi l’andamento di una reazione chimica.

 The course intends to reach two different goals depending on the Synthesis and the Mechanism section.

Synthesis Section (Teacher Prandi)

At the end of the course, the student  will be able to choose the method best suited for the formation of carbon-carbon bonds using organometallic compounds (prepared in advance or generated in situ), p complexes of transition metals, with the maximum control of regioselectivity, chemoselectivity and stereoselectivity.

 Part "Mechanisms"  (G. Tonachini):

The purpose is to provide simple theoretical tools, suitable to rationalize or predict qualitatively the outcome of some chemical reactions.

Per la parte di Sintesi (Docente Prandi): Il corso si propone di ampliare ed approfondire la conoscenza nel campo della sintesi e della reattività di strutture molecolari organiche attraverso l’apprendimento di concetti e metodi che caratterizzano la sintesi organica moderna. L’allievo dovrà essere in grado di affrontare con spirito critico la sequenza di passaggi proposti per una  sintesi organica, fornendo anche suggerimenti personali al fine di ottimizzare i risultati voluti; di valutare gli aspetti chemo-, regio- e stereoselettivi di alcune importanti reazioni.

Parte "Meccanismi" (G. Tonachini):

Mettere in grado lo studente di elaborare una razionalizzazione dei motivi per cui una reazione organica è comune oppure non osservata. Lo studente dovrebbe essere in grado di definire una reazione come “permessa” o “proibita”, intendendosi che il suo decorso sia relativamente facile oppure che presenti una barriera di energia significativa di origine elettronica. Questa capacità può essere introduttiva a studi ulteriori che mirino ad un esame quantitativo del decorso di una reazione chimica (energetica del processo, termochimica, valutazione quantitativa del meccanismo preferito, rapporto con la cinetica).

 

 Synthesis Section (Teacher Prandi)

The course aims to broaden and deepen the knowledge in the field of synthesis and reactivity of organic molecular structures by learning concepts and methods that characterize the modern organic synthesis. The pupil must be able to deal with a critical sequence of steps proposed for an organic synthesis, as well as providing personal suggestions in order to optimize the desired results; to evaluate the aspects chemo-, regio- and stereoselective of some important reactions.

 Part "Mechanisms"  (G. Tonachini):

The objective is to make the student capable of rationalizing the reasons why an organic reaction is commonly observed or not. The student should acquire the capability of defining a reaction as “allowed” or “forbidden”, meaning by these words that it is either energetically easy or presents a sizable barrier of electronic origin. This capability can be introductory to further learning aiming to define quantitatively the evolution of a chemical system (energetics and thermochemistry, mechanism assessment, relation with kinetics).

Tradizionale: lezioni frontali in aula con l'ausilio di materiale PowePoint  messo a disposizione anticipatamente sul sito web del corso.

Parte "Meccanismi" (G. Tonachini):

Il modulo consta di 3 CFU, equivalenti a 24 h di lezione. Le lezioni sono svolte principalmente alla lavagna, con l’ausilio del colore nel tracciare diagrammi e disegnare strutture molecolari.

Lessons (3 credits, i.e. 24 hours) , delivered mainly by writing on the blackboard, with color drawing of molecular structures and diagrams.

Modalità di verifica dell'apprendimento

Parte "Sintesi" (C. Prandi)

L' esame prevede  una prova orale che consiste in tre domande alle quali lo studente dovra rispondere dimostrando di aver compreso i meccanismi delle reazioni trattate e progettare brevi sintesi totali.

Parte "Meccanismi" (G. Tonachini):

L’esame orale comprende da parte dello studente, oltre all’esposizione discorsiva sul tema richiesto, la stesura di diagrammi di vario tipo e il disegno di orbitali molecolari (MO) interagenti. L’esame è teso a verificare la chiarezza di idee e la chiarezza espositiva con cui lo studente usa speditamente i due strumenti teorici qualitativi (“di pronto impiego”) proposti nel corso d’insegnamento. Con essi si devono esaminare una o due reazioni scelte tra una varietà di reazioni organiche, con lo scopo di definire se essa sia facile e veloce oppure lenta e magari non osservata, e inoltre magari di spiegare varie forme di selettività. L’esame orale permette un dialogo che pare possa evidenziare (più facilmente di uno scritto) il buon uso della logica e la coerenza nell’elaborare le analisi richieste.

Lo studente può scegliere liberamente l'ordine con cui sostenere le due parti, con il vincolo temporale di 6 mesi e/o tre appelli tra di esse, passato questo intervallo il voto già acquisito non sarà più considerato valido. Il voto finale è la media matematica degli esiti delle prove sostenute con i due docenti.

 Part "Synthesis" (C. Prandi)

The oral exam is based on three different questions related to the three parts of the course. The student is required to demonstrate his knowledge on reaction mechnisms and desigh simple synthetic sequences.

 Part "Mechanisms"  (G. Tonachini):

The oral examination encompasses both a discussion of the requested theme and the drawing of diagrams of different kind and sketches of interacting molecular orbitals (MOs). It aims to assess how clear are the concepts to the student, and how lucidly she/he can talk about and use them. The two qualitative theoretical methods offered in the lectures should be used promptly by applying them to one/two reactions selected among a variety of organic reactions. The target is defining if a reaction is easy and fast, or slow and perhaps not observed. Moreover various forms of selectivity might be discussed. The oral examination is believed to allow the assessment of the logic capabilities of the student, more than a written test.

The student can choose the order of the examination, but the delay between the exams of the two parts can not be longer than  6 mounths . The mark is the average of the two outcomes.

SINTESI ENANTIOSLETTIVA. Definizioni. Strategie sintetiche, chiral pool, catalizzatori chirali, reagenti chirali

REAZIONI DI CROSS-COUPLINGS. Generalità sui cross-couplings catalizzati dal palladio(0): reazioni di Heck, Suzuki, Stille, Negishi, Sonogashira e loro aspetti chemo e stereochimici. Applicazioni principali in sintesi con esempi. Complessi π-allilici del Pd. Reazioni metallo-ene. Reazioni domino. Reazioni catalizzate da Pd(II). Il ruolo dei leganti, angoli di cono, fosfine bidentate, fosfine di Buchwald, NHC carbeni. Reazione di Buchwald-Hartwig.

Catalisi da ORO. Reazioni di enini, attivazione di legami multipli nei confronti di nucleofili. Catalisi eterogenea, l' ro nelle reazioni di cross-coupling.

  • Teoria degli Orbitali di Frontiera e Regole di Woodward e Hoffmann. Concetti generali.
  • Effetti sulla cinetica delle energie degli orbitali molecolari (MO).
  • Effetto della loro polarizzazione sulla orientazione (regiochimica).
  • Come gli MO possano influire sulla diastereoselettività.
  • Applicazione a reazioni di cicloaddizione, reazioni elettrocicliche, reazioni chelotropiche, migrazioni sigmatropiche. Reazioni ioniche, radicaliche e fotochimiche.
  • Diagrammi di correlazione di MO, di configurazioni elettroniche, di stati elettronici. Incroci reali ed evitati. Esempi.
  • Transizioni diabatiche, relazione di Landau-Zener.

Chemoselectivity and Diasteroselectivity

ENANTIOSELECTIVE REACTIONS. Definitions, synthetic strategies, chiral pool, chiral catalysts, chiral reagents. Examples in asymmetric syntheses.

CROSS-COUPLINGS REACTIONS. General information on cross-couplings catalyzed by palladium (0).  Heck, Suzuki, Stille, Negishi, Sonogashira reactions and their chemo and stereochemical aspects. Main applications in synthesis with examples. Π-allyl complexes of Palladium. Metallo-ene reactions. Domino reactions. Reactions catalyzed by Pd (II). The role of the ligands, cone angles, bidentate phosphines, Buchwald phosphines, NHC carbenes. Buchwald-Hartwig reaction.

Catalysis by GOLD. Reactions of enynes, activation of multiple bonds towards nucleophiles. Heterogeneous catalysis, gold in cross-coupling reactions.

 

  • Frontier Orbitals Theory and Woodward-Hoffmann rules. General concepts.
  • How MO energies can affect reaction rates.
  • How their polarization can determine regioselectivity effects.
  • How they can affect the diastereoselectivity.
  • Application to cicloadditions, cheletropic reactions, electrocyclic reactions, sigmatropic rearrangements.
  • Correlation diagrams for MOs, electron configurations, and electronic states. Real and avoided crossings. Exemples.
  • Diabatic transitions, Landau-Zener equation.
