Obiettivi formativi

L’insegnamento concorre alla realizzazione degli obiettivi formativi del Corso di Laurea Magistrale in Chimica ponendo l'attenzione sulle macromolecole in termini di caratterizzazione strutturale e attività biologica per l’utilizzo finale in campi come la salute e la green chemistry.

In particolare, il corso fornisce allo studente conoscenze dettagliate su:

- relazione struttura-funzione delle macromolecole biologiche

- studio del folding delle proteine

- evoluzione di strutture proteiche e di moduli proteici

- metodi spettroscopici per lo studio della struttura delle proteine

- cristallografia ai raggi X

- analisi e costruzione di modelli molecolari di strutture proteiche

- Attivazione di ossigeno molecolare da cofattori presenti nelle proteine

- analisi di struttura-funzione applicata a emo-proteine, flavo-proteine e rame-proteine.

Nelle esercitazioni di laboratorio lo studente imparerà a generare modelli di strutture proteiche con approcci bioinformatici e utilizzerà metodi sperimentali di cristallizzazione delle proteine.

The course contributes to the objectives of the Master Degree Course in Chemistry, focusing on macromolecules in terms of structural characterization and biological activity for end use in fields such as health and green chemistry.

In particular, the course provides the student with detailed knowledge of:

- Structure-function relationships of biological macromolecules

- protein folding

- Evolution of protein structures and protein modules

- Spectroscopic techniques applied to the study of protein structure

- X-ray crystallography applied to proteins

- Analysis and construction of molecular models of protein structures

- Activation of molecular oxygen by cofactors found in proteins.

- Analysis of structure-function applied to haem-, flavo-proteins and copper proteins.

In the practicals, the student will learn to create models of protein structures through bioinformatic approaches and will use experimental methods for protein crystallization

CONOSCENZA E COMPRENSIONE

Al termine dell’insegnamento lo studente comprenderà:

- le differenze tra elementi di struttura primaria, secondaria e terziaria e come questi diversi livelli di organizzazione possono essere studiati con diverse metodologie spettroscopiche.

- i principi e i metodi per la cristallografia a raggi X applicata a proteine.  

- come la struttura 3D di una proteina influenza la sua funzione.

- perché l'ossigeno molecolare deve essere attivato da diversi enzimi e quali cofattori contribuiscono a questa attivazione.

Nella parte delle esercitazioni, lo studente imparerà a generare da una sequenza primaria di una proteina un modello 3D in silico. Inoltre, avrà un'esperienza diretta di cristallizzazione delle proteine utilizzando il lisozima. Durante il corso gli studenti inoltre hanno la possibilità di presentare un recente articolo della letteratura in lingua inglese evidenziando anche la capacità di lavorare in gruppo

CAPACITÀ DI APPLICARE CONOSCENZA E COMPRENSIONE 

Al termine dell'insegnamento, lo studente sarà in grado di :

- Riconoscere e classificare le struttue proteiche.

- Comprendere come gli enzimi attivano l'ossigeno molecolare per applicazioni biocatalitiche che includono il metabolismo di farmaci e la green chemistry.

- Selezionare la tecnica spettroscopica appropriate per studi strutturali e funzionali di proteine.

- Visualizzare le strutture in 3D con software appropriati

- Utilizzare databases di sequenze e strutture proteiche

 

KNOWLEDGE & UNDERSTANDING

At the end of the course students will understand:

- the differences between elements of primary, secondary and tertiary structure;

- how these different levels of organization can be studied with spectroscopic techniques.

- principles and methods for X-ray crystallography applied to proteins.

- how the 3D structure of a protein influences its function,

- why molecular oxygen needs to be activated by different enzymes within bacteria and human and which cofactors contribute to this activation.

In the practicals, students will learn how to generate a protein 3D model in silico starting from a primary sequence. Moreover, the students will have direct experience of protein crystallization using lysozyme. During the course the students also get a chance to present a recent literature article in English highlighting their ability to work in teams.

APPLYING KNOWLEDGE &UNDERSTANDING

At the end of the course, the student will be able to:

- Recognize and classify protein structures.

- Understand how enzymes activate molecular oxygen for biocatalytic applications that include drug metabolism and green chemistry.

- Select the appropriate spectroscopic technique for structural and functional protein studies.

- View 3D structures with appropriate software

- Use databases of sequences and protein structures

La frequenza alle lezioni è facoltativa, mentre al laboratorio è obbligatoria

Lecture attendance is optional, while practicals is compulsory.

Modalità di verifica dell'apprendimento

Prova scritta finale unica con 3 domande (80% voto finale), di cui due domande sul modulo della Prof. Sadeghi e una sul modulo della Dr. Di Nardo.

Ogni domanda ha un peso di 10 punti.

La relazione scritta sulla parte esercitativa di bioinformatica (modeling) viene valutata (10% del voto finale).

I seminari degli studenti vengono anche valutati (10% del voto finale).

Il voto finale è espresso in 30esimi.

La commissione d’esame si riserva di avere una discussione dell’elaborato al momento della registrazione del voto.

Final written exam with 3 questions (80%), two of them about the module of Prof. Sadeghi, one of them about the module of Dr. Di Nardo.

Each question is 10/30.

The written report on thebioinformatics (modeling) is evaluated ( 10 % of the final grade ).

The seminars of the students are also assessed ( 10 % of the final grade ).

 The final grade will be expressed in a maximum of 30.

The examination committee reserves the right to have an oral exam to clarify the preparation of the student before registering to vote.
