Obiettivi formativi

L‘insegnamento si propone di fornire a uno studente della LM in Chimica le basi per una sua futura collocazione professionale Ricerca/Sviluppo in ambito Farmaceutico (Drug Design/Development). Mantenendo la molecola organica come punto centrale dell’insegnamento, lo studente sarà gradualmente introdotto al linguaggio e alle metodologie che un chimico utilizza nell‘affrontare le varie problematiche in cui è coinvolto in ambito farmaceutico.

Partendo da un‘introduzione ai principi generali della Chimica Farmaceutica (Medicinal Chemistry), il corso approfondirà due temi generali: Sintesi Farmaceutica (adattamento delle metodologie di sintesi avanzata all‘ambito farmaceutico) e Sviluppo Farmaceutico (tecniche di ottimizzazione del processo sintetico portandolo dal laboratorio all‘ impianto pilota).

The course aims to provide a student of Chemistry LM basis for his future professional placement in Research / Development within of Drug Design / Development areas. Maintaining the organic molecule as the central point of the course, students will be gradually introduced to the language and methodologies that a Chemist used in addressing the various issues that are involved in the pharmaceutical field.

Starting with an introduction to the general principles of Medicinal Chemistry, the course will explore two general themes: Pharmaceutical Synthesis (adaptation of synthesis methods advanced scope pharmaceutical) and Pharmaceutical Development (technical optimization of the synthetic process by bringing the lab to pilot plant then to large scale production).

AI termine dell'insegnamento, lo studente dovrà dimostrare di essere in grado di applicare i principi fondamentali della Chimica Farmaceutica al design di molecole bioattive. Lo studente dovrà essere inizialmente in grado di giudicare la possibilità che una molecola organica possa considerarsi un potenziale farmaco (profilo drug-like) prevedendo la presenza di eventuali debolezze (solubilità, biodisponibilità, stabilità metabolica, ….). La molecola bersaglio dovrà essere quindi trattata in termini di sintesi farmaceutica e quindi di sviluppo farmaceutico, passi fondamentali per portare una struttura bersaglio dalla cappa del Chimico fino alla sua produzione in ampia scala. 

At the end of the course, students will have to prove the acquisition of knowledge of the principles of Medicinal Chemistry in the design of bioactive molecules. The student will initially be able to judge the possibility that an organic molecule can be considered as a potential drug (drug-like profile) providing for possible weaknesses (solubility, bioavailability, metabolic stability, ....). The target molecule will then be treated in terms of pharmaceutical synthesis and then pharmaceutical development, basic steps to bring a target structure from the hood of Chemical until its production in large scale.

Modalità di verifica dell'apprendimento

L'esame consiste in una prima prova, scritta, il cui superamento permette l‘accesso a una seconda prova, questa volta orale. Nella determinazione del voto finale viene tenuto conto di entrambe le prove.

Prova scritta. Allo studente è fornito un dettagliato profilo descrittivo riferito a una struttura organica incognita. La struttura è descritta per le sue proprietà chimico-fisiche, (solubilità, logP, …), per profilo druglike quindi per reattività, debolezze strutturali e possibile metabolismo. Lo studente è chiamato a scegliere fra 20 possibili strutture la struttura  che risponde al profilo fornito, motivando le proprie decisioni.

Prova orale. Circa 48 ore prima dell’ esame, lo studente riceve un articolo di recente pubblicazione in campo MedChem. Simulando un group meeting in ambito di Ricerca, lo studente preparerà la presentazione orale in PowerPoint di 10 min durante la quale descriverà alcune molecole presentate nel lavoro. Durante la presentazione, tali molecole saranno dettagliatamente discusse secondo le tematiche del Corso e forniranno occasioni per nuovi spunti di discussione (10 min).

The final examination consists of a first test, written-test, which allows the access to a second test, this time oral. In determining the final grade is given by both tests.

Written test. The student is provided a detailed descriptive profile refers to an organic structure unknown. The structure is described for its physical-chemical properties (solubility, logP, ...), for profile druglike then for reactivity, structural weaknesses and possible metabolism. The student is asked to choose among 20 possible structures  the candidate able to responds to the profile provided, giving reasons for its decisions.

Oral examination. About 48 hours before the oral examination, the student receives an article recently published in the field medchem. Simulating a group meeting in the area of ​​search, the student will prepare the oral presentation in PowerPoint 10 min during which describe molecules presented in the work. During the presentation, such molecules will be discussed in detail under the themes of the course and will provide opportunities to explore new areas for discussion (10 min).

1) The Modern Drug Discovery Process.Nella parte iniziale dell’ insegnamento sarà descritta nel dettaglio la serie di passaggi che vengono seguiti nel moderno Drug Design  per portare una molecola ad attività in vitro fino alla commercializzazione sul mercato quale nuovo Farmaco. La conoscenza delle varie fasi servirà successivamente per approfondire le singole problematiche e legarle alla struttura molecolare, soggetto dell’ insegnamento.

2) Correlation between structural features and chemiophysical properties: a trip through the body.Focalizzandosi quindi sulla struttura molecolare, in questo momento saranno approfondite le correlazioni fra scaffold molecolare (gruppi funzionali) e il profilo chimico fisico. L’obiettivo è rendere lo studente in grado di discutere la correlazione di descrittori molecolari quali solubilità, log P, pKa con le caratteristiche strutturali. La struttura verrà quindi descritta in termini di reattività/debolezze strutturali. Questa fase, prettamente chimica, sarà fondamentale per poter poi muovere gli stessi concetti a livello biologico.Qui le capacità della struttura molecolare di interagire con il suo intorno determineranno la sua vita nel sistema biologico. Il candidato Farmaco dovrà quindi essere ottimizzato per ogni fase della sua vita, dalla somministrazione, all’arrivo sul sito bersaglio quindi alla generazione dell’effetto farmacologico e poi all’escrezione (Metabolismo). In questa fase verranno anche citate nozioni di base di Tecnica Farmaceutica.

3) The Drug Discovery process: the role of an Organic Chemist. Come si colloca un Chimico Organico in un processo di Drug Discovery? Quali problematiche specifiche viene chiamato a risolvere? In questa fase si cercherà di rispondere a queste due domande introducendo il concetto di Pharmaceutical synthesis, un’ideale branca della sintesi organica che accorpa tutte le metodologie atte ad accelerare il processo sintetico muovendolo ad ampi numeri. Partendo da esempi di sintesi totali fondamentali (Chinina, Taxol) saranno introdotti i concetti di Hit-to-Lead optimization, Combinatorial Chemistry, Parallel synthesis, Click chemistry, Solid phase synthesis, Scavenging reagents e Flow Chemistry.

Case study:             Discovery of antimalarial drugs: from Quinine to Artemisinin and beyond.

Case study:             Cytotoxic Drugs: the total  synthesis of Taxol (Paclitaxel).

4) From Research to Development: the role of an Organic Chemist. Una struttura molecolare dopo essere stata ottimizzata per attività, tossicità e ADME si trova a subire ancora un ultimo, fondamentale, passaggio prima di poter arrivare sul mercato. In questo ultimo capitolo viene descritto il cosiddetto Sviluppo (Development) il passaggio che porta la metodologia sintetica sviluppata per bassa scala (all’ interno del grammo) ad essere condotta su scala Pilota prima e poi su impianto vero e proprio. A chiusura del capitolo verranno trattati anche aspetti normativi.

1) The Modern Drug Discovery Process. In the initial part the course, it will be described in detail the steps that are followed in the modern drug design to bring a molecule with in vitro activity to commercialization on the market as a new drug. The knowledge of the various phases will the entry-level knowledge necessary for a deeper treatment of each individual topic.

 

2) Correlation between structural features and chemiophysical properties: a trip through the body. Focusing on the molecular structure, at first will be explored the correlations between molecular scaffold (functional groups) and a physical-chemical properties. The goal will be to make students able to discuss the correlation of molecular descriptors such as solubility, log P, pKa with structural features. The molecular structure is then described in terms of reactivity / structural weaknesses. This phase, where a lot of chemistry is involved, will be crucial in the further step, when the same concepts are moved at biological level. The ability of the molecular structure to interact with its surroundings will determine his life in the biological system. The candidate drug will then be optimized for each phase of his life, from the administration, the arrival on the target site then the generation of the pharmacological response and then excretion (Metabolism). In this phase we will be also mentioned basics of Pharmaceutical Technology.

3) The Drug Discovery process: the role of an Organic Chemist.How can an Organic Chemistry professional figure can be involved in a Drug Discovery process? What specific problems he/she is called to solve? In this phase, we will try to answer these two questions by introducing the concept of Pharmaceutical synthesis, an ideal branch of organic synthesis that brings together all methods designed to speed up the process by moving to large numbers synthetic. Starting with examples of total syntheses fundamental (Quinine, Taxol) will be introduced to the concepts of:

Hit-to-Lead optimization, Combinatorial Chemistry, Parallel synthesis, Click chemistry, Solid phase synthesis, Scavenging reagents e Flow Chemistry.

4) From Research to Development: the role of an Organic Chemist.A molecular structure, after being optimized in terms of activity, toxicity and ADME, has to undergo one last, crucial step before it can reach the Market. In this last chapter describes the so-called Development, the passage that carry the synthetic methodology, developed for low-scale (inside the gram scale), to be conducted on scale Pilot first and then on the lerga scale production. At the end of the chapter, also regulatory issues will be discussed.
